Leukopaks are highly-enriched leukapheresis-derived products obtained from an IRB-approved collection facility. Leukopaks contain higher concentrations of mononuclear cells, B cells, T cells, stem/progenitor cells, dendritic cells, and other cell types as compared to standard venipuncture collection methods or buffy coat products.
Leukapheresis is performed using the Spectra Optia® Apheresis System with healthy, consented donors following IRB-approved protocols. MNC rich cells are continuously collected from 1-1.5 donor blood volumes into a sterile bag using ACD-A as an anticoagulant. After a sample is withdrawn for quality testing, a nominal volume of 145mL is transferred into a sterile transfer bag prior to shipping.
These products are transferred into a sterile blood bag to contain a nominal, calculated volume of a collection, regardless of the cell count. Approximately 60mL of autologous plasma is automatically collected directly into the cell pool following leukapheresis. The total nucleated cell count is determined using AO/PI from the collection and provided as information. An average half procedure produces ~8.75B TNCs with >90% of collections having 6B TNCs or more at release. These products are released based on the volume of apheresed material provided.
The actual TNC concentration and cell total count will vary from collection to collection. Apheresed total nucleated cell (TNC) products typically contain up to 50% T cells, 20% monocytes, 10% B cells, and 10% NK cells, 3% granulocytes, and 3% hematocrit.
