The NucleofectorDevice is the single cuvette based system that has been used in research labs since 2001. It allows efficient transfection of hard-to-transfect cell lines and primary cells with different substrates (e.g., DNA vectors or siRNA oligonucleotides) in low-throughput format. The NucleofectorII/2b Device can also be used for bacteria transformation by using alternative cuvettes (for ordering information see below). If you are not sure about the device version of your NucleofectorII please check the rear panel and firmware version of your device (see descriptions below). To view the current firmware and program software version of your NucleofectorII or IIS, turn on the Nucleofectorby using the power button. If the NucleofectorII Device starts in the »Free Program Choice« mode, exit this mode by pressing the »X« button. Press the functional button »menu« several times until »Software Versions« appears. The first set of digits encodes the firmware version. The third (NucleofectorII: S3-x) or second set of digits (NucleofectorIIS: S4-x) encodes the program software version. NucleofectorII Rear panel of device without receptacles for USB and serial connection Current program software version number S3-8 NOT compatible with SCN NucleofectorKits. If your NucleofectorII has an older program software version please order the respective chipcard (see link in the left navigation) and refer to the upgrade instructions for NucleofectorII. NucleofectorIIS Rear panel of device provides receptacles for USB and serial connection. Firmware version number N5-5 or higher. Current program version number S4-5. Compatible with SCN NucleofectorKits 4D-NucleofectorConsumables The 4D-NucleofectorSystem can handle four different NucleocuvetteVessels all using our innovative conductive polymer electrode material. For Nucleofectionin suspension (X Unit): 100 µl NucleocuvetteVessels: These cuvettes hold 100 µl of cell suspension and are perfectly suited for medium to high cell numbers (105 to 107). 16-well NucleocuvetteStrips: This vessel allows convenient transfection of up to 16 samples in parallel. The volume of 20 µl per cavity is designed for low to medium cell numbers (104 to 105). For Nucleofectionin adherence (Y Unit):24-well Dipping electrodes: For transfection, an array of 24 dipping electrodes is inserted into a 24-well culture plate containing the adherent cells. For more details please see our adherent Nucleofectionpage.Same Protocols for Various Cell Numbers The use of conductive polymer for all our NucleofectionVessels allows seamless transfer of protocols from 20 µl NucleocuvetteStrips to 100 µl Nucleocuvettesand vice versa. Thus no tedious re-optimization is required. You can simply use the same conditions for both volumes. Simplified Kit Concept for Primary Cells Due to the unique characteristics of our conductive polymer material we were able to streamline our kit concept for primary cells. For the 4D-NucleofectorSystem only five different Primary Cell NucleofectorKits are required to cover a broad range of primary cell transfection challenges. Current program software version is V2.5 If this number is lower than the current number, you can order the respective chipcard (see link in the left navigation) and update the programs according to the upgrade instructions for the NucleofectorI.
